Cloning and Characterization of a Citrate Sythase Gene from Bdellovibrio bacteriovorus

In terms of carbon utilization and ATP production, Bdellovibrio has one of the highest energy efficiencies of any bacterium. Bdellovibrio is also known to possess elevated citric acid cycle enzymes. However, it is not known whether Bdellovibrio can survive using alternative forms of catabolic pathways to produce energy. Via allelic exchange of inactivated citrate synthase genes, I hope to determine if the citric acid cycle is a necessary catabolic pathway or a preference over anaerobic or less efficient channels. I plan to accomplish this task by investigating the role of the citric acid cycle and the corresponding regulatory genes and enzymes. Once understood, my goal to gain a better understanding of Bdellovibrio bacteriovorus lifecycle will hopefully be realized.
